Custom Plasmids

The starting point for plasmid construction is rarely the same from one project to another. That is why we have developed an approach with no prerequisites: you provide whatever material or information you have, and we take care of the rest.

Starting material may take many forms: a coding or regulatory nucleic acid sequence, a protein sequence, a GenBank or UniProt identifier, or simply a bibliographic reference. In the latter case, our experts identify and retrieve the sequence of interest on your behalf.

Several vector options are available: we can select the backbone best suited to your expression system from our in-house collection, or work with your own acceptor vector under strict confidentiality. You may also provide us with a plasmid sequence that you do not physically possess, and we will reconstruct it.

The diversity of the constructs we produce—including large inserts, high-GC sequences, repeated elements, and toxic fragments—demonstrates our command of a broad range of cloning strategies, selected and adapted to the specific requirements of each project.

RD-Biotech adapts to your intellectual property requirements. For performance-driven projects, we use licensed technology that delivers transient antibody-expression yields of up to 1 g/L; in this model, the plasmid material remains the property of RD-Biotech.

For projects requiring technology transfer, we work with royalty-free vectors that are made fully available to the client upon project completion.

Plasmids for mRNA production by in vitro transcription

The quality of mRNA produced by in vitro transcription depends first and foremost on the quality of its plasmid template. The transcription promoter, 5′ and 3′ untranslated regions (UTRs), open reading frame (ORF), poly(A) tail, and linearization site all directly influence transcription yield, mRNA stability, and translation efficiency.

RD-Biotech can contribute at every stage of the design process. We can modify an existing plasmid by:

  • changing the ORF, optimizing the UTRs, adjusting poly(A)-tail length, and repositioning the linearization site; or
  • building a new template from royalty-free vectors into which we incorporate royalty-free UTRs combined with the ORF of your choice.

Plasmid linearization can also be performed in-house, as can the production of small IVT mRNA batches for proof-of-concept (POC) validation.

Plasmids for viral vector production and gene therapy

Viral vector production relies on a multiple-plasmid system whose quality and design directly determine the performance of the final vector. RD-Biotech has expertise in constructing all of these components—helper, packaging, and transfer plasmids—for the main systems used in gene therapy, primarily AAV and lentiviral vectors, with other systems available on request.

The transfer plasmid lies at the heart of our molecular biology work. It carries the transgene of interest and its associated regulatory elements—including the promoter, ITRs, packaging signals, and insulator elements. Its design can be fully customized according to your therapeutic target, AAV serotype, cell tropism, or insert-capacity constraints.
