Every stage is optimized to maximize yield and purity.

  • Production in 250 mL to 15 L shake flasks
  • Production in 250 mL to 10 L fermenters
  • Collection of bacterial pellets by centrifugation
  • Solubilization of bacterial pellets using sonication, microfluidization, French press processing or various detergents and denaturing agents, including urea and guanidine
  • Affinity chromatography using Ni-NTA

The harvested culture supernatant can be processed by tangential flow filtration and concentrated up to 20-fold. This step optimizes purification for large production volumes.

The concentrated culture supernatant can also be supplied as the finished product, with optional diafiltration into PBS buffer.

  • Tag-free: Vector design incorporating an enzymatic cleavage site (TEV, Enterokinase, SUMO, PreScission, etc.), followed by complete cleavage and a separation step to yield a native recombinant protein free of tag residues. This option is particularly valued for structural studies, functional activity assays, and preclinical applications.
